The involvement of IRAK-1 in the regulation of NFATc2 in T cells

چکیده
Interleukin-1 receptor associated kinase -1 is a protein kinase pivotal in mediating signals for innate immune responses. Here, I report that IRAK-1 also regulates cell-mediated immune responses. NFATc2 (nuclear factor of activated T cells) was found to be associated with IRAK-1 in T cells in vitro and its activity was elevated in the absence of IRAK-1. In addition, IRAK-1 -/mice had increased naturally occurring regulatory T cells and inducible regulatory T cells as well as Th1 responses as compared to WT mice. The findings suggest that activated T cells might employ IRAK-1 to mediate the regulation of acquired immunity. Therefore, IRAK-1 may participate in direct signaling cross talk between the innate and the acquired immunity.

Differential regulation of Foxp3 and IL-17 expression in CD4 T helper cells by IRAK-1.
Host immune responses are finely regulated by the opposing effects of Th17 and T regulatory (Treg) cells. Treg cells help to dampen inflammatory processes and Th17 cells facilitate various aspects of immune activation. The differentiation of Th cells depends on a unique combination of stimulants and subsequent activation of diverse transcription factors. In particular, cooperative activation of...

The nuclear factor of activated T (NFAT) cell family of transcription factors is important in regulating the expression of a broad array of genes, including cytokines, T cell surface receptors, and other transcription factors. NFATc1 and NFATc2 are two principal NFAT members that are expressed in peripheral T cells. Levels of NFAT expression in T cells are partly transcriptionally regulated, bu...
متن کاملInhibition of Interleukin-1 Receptor-Associated Kinases 1/4, Increases Gene Expression and Serum Level of Adiponectin in Mouse Model of Insulin Resistance
Insulin resistance is a feature of most patients with type 2 diabetes mellitus. Epidemiological evidence suggests a correlation between inflammation and insulin resistant states such as obesity, but the underlying mechanisms are largely unknown. Interleukin-1 receptor-associated kinases (IRAK) play a central role in inflammatory responses by regulating the expression of various inflammatory gen...
متن کاملThe NFATc2 gene is involved in a novel cloned translocation in a Ewing sarcoma variant that couples its function in immunology to oncology.
PURPOSE Ewing sarcoma is an aggressive sarcoma and is the second most common bone sarcoma in childhood. Disease-specific t(11;22) ( approximately 85-90%), t(21;22) ( approximately 5-10%), or rarer variant translocations with the involvement of chromosome 22 ( approximately 5%) are present. At the gene level, the EWSR1 gene fuses with FLI1, ERG, or other ETS transcription factor family members. ...
متن کاملFocal Adhesion Kinase (FAK) Involvement in Human Endometrial Remodeling During the Menstrual Cycle
Background: Endometrial remodeling occurs during each menstrual cycle in women. Reports have shown that, in a variety of cell types, processes such as proliferation, signaling complex formation and extra cellular matrix remodeling require a cytoplasmic tyrosine kinase, focal adhesion kinase (FAK). The present study has focused on the expression pattern of FAK in human endometrium during the men...
